Purification of chlorogenic acid in extracts of waste and inferior tobacco leaves

The effects of experimental conditions, such as injected solution concentration, concentration and volume of elution solution, for column chromatography, and sample solution’s pH value, extracting time and number of repetitions on the purification of chlorogenic acid in extracts of waste and inferior tobacco were investigated. The results showed that: 1) the proper conditions of column (glass column 16 mm×200 mm, bed volume 19.8 mL) chromatography by polyamide resin were: concentration of crude chlorogenic acid solution 8.5 mg/mL, elution solution 198.0 mL 25% ethanol; 2) the appropriate conditions of extraction with ethyl acetate were: pH value of sample solution 2.0, extracting twice, extraction time 5 minutes. Under the mentioned conditions, the content of chlorogenic acid in the obtained product reached 82.1% with the yield of 57.6%.
